I assume that you have directions to Anthony's, but if I may spice it up a bit without adding time, come down Guelph Line from the 401. It's a lot more fun to drive. Once you start coming down the escarpment, you will get to Dundas (Hwy 5) turn left/ east on Dundas. Seven lights later (I think) is Appelby. Turn right/south on Appelby. Once you go under the railroad tracks turn left at the street light. You'll see us up on the left.

fastest route would be this one from map quest from the rest stop Mc Donald's on the 401 East
1: Start out going EAST on PROVINCIAL ROUTE 401 E. 20.5 miles
2: Take EXIT 312 toward RR-1 / GUELPH LINE / CAMPBELLVILLE / BURLINGTON. 0.2 miles
3: Turn LEFT onto REID SDRD. <0.1 miles
4: Turn RIGHT onto MAIN ST N / RR-1 / RR-9. Continue to follow RR-1. 4.5 miles
5: Turn LEFT onto DERRY RD / RR-7. 1.7 miles
6: Turn RIGHT onto APPLEBY LINE. 7.4 miles
7: Turn LEFT onto DRYDEN AVE. 0.4 miles
8: End at 5173 Dryden Avenue

But personally I like using highway 6 as it's more faster as the lanes are larger so it would be like this:
1: 401 east from the Mc Donald's rest stop
2: Exit at Hwy 6 South to Hamilton
3: enter Highway 403 east from Highway 6
4: exit off at Appleby Line exit
5: Turn North on to Appleby Line
6: Turn Right onto DRYDEN AVE.
